چکیده

The present study is concerned with the kinetics and thermodynamic study of urease extracted from soybeans. The effect of pH, temperature, concentration of substrate and incubation time on the urease activity was examined. Maximum urease activity (86.79 ± 2.43 U/g) was obtained at 30 o C and pH 8.0 after 60 sec of incubation. Kinetics constants i.e. Km and Vmax were estimated by using Lineweaver-Burk plot and were found to be 36.49 mM and 87.72 U/g, respectively. Thermodynamic constants i.e. Activation energy (Ea), change in enthalpy (ΔH) and change in entropy (ΔS) were calculated using Arrhenius plot and were found to be 5.7 KJ/mol, 3.468 KJ/mol and -4.57 KJ/mol respectively.

A Thermodynamic Study of the Interaction between Urease and Copper Ions

A thermodynamic study of copper ions by jack bean urease (JBU) was carried out at two temperatures of 27 and 37?C in Tris buffer (30 mM; pH=7.0) using an isothermal titration calorimetry. There is a set of twelve identical and non-interacting binding sites for copper ions. The intrinsic dissociation equilibrium constant and the molar enthalpy of binding are 285 µM and ?15.2 kJ/mol at 27?C and 3...

Cadmium removal from wastewater using nano-clay/TiO2 composite: kinetics, equilibrium and thermodynamic study

In this research, commercial nano-clay (NC) was modified with TiO2 functional groups and characterized via XRD and FTIR methods. The modified nano-clay was applied as an adsorbent for the removal of cadmium from wastewater solutions. The effects of the operating parameters including initial pH, cadmium concentration and adsorbent concentration were analyzed by the Taguchi method. The...

Effects of dry matter content on trypsin inhibitors and urease activity in heat treated soya beans fed to weaned piglets

A nutrition study was conducted to evaluate the growth response of weaned piglets fed diets containing soya beans that had been processed into protein supplements at two different levels of dry matter (DM) and temperature. Four diets contained protein supplements prepared from whole full-fat soya beans equilibrated at 800 or 900 g kgÿ1 DM prior to being heated to 110 or 1258C. An additional die...
